目的研究虾青素对人胃癌细胞SGC-7901增殖和凋亡的影响。方法采用体外培养、细胞增殖实验,以可见光、荧光、激光扫描共聚焦显微镜(LSCM)观察胃癌细胞的形态变化。结果虾青素对SGC-7901细胞增殖具有抑制作用,IC50为2.5mmol/L;分别用含0.42、0.84、1.68mmol/L的虾青素的培养介质处理SGC-7901细胞48h,在可见光、荧光显微镜、LSCM下观察细胞形态,分别出现了细胞数量明显减少、皱缩变形、体积缩小、触角伸长、细胞表面凸起多个小泡,显示亮黄色荧光的细胞核呈现“新月状”、条状甚至碎片状形状,凋亡小体,DNA面积较明显减小等典型的凋亡细胞形态特征。结论虾青素对SGC-7901细胞的增殖具有明显的抑制作用,且诱导胃癌细胞凋亡。
Objective To investigate the effects of astaxanthin on the proliferation and apoptosis of human gastric cancer cell line SGC-7901. Methods The morphological changes of gastric cancer cells were observed by visible light, fluorescence and laser scanning confocal microscopy (LSCM) in vitro. Results Astaxanthin inhibited the proliferation of SGC-7901 cells with an IC50 of 2.5 mmol / L. SGC-7901 cells were treated with astaxanthin containing 0.42, 0.84 and 1.68 mmol / L, respectively, for 48 h. Under visible light and fluorescence Under microscope and LSCM, the morphological changes of cells were observed. The numbers of cells were significantly decreased, the cells were shrunk, the volume was reduced and the antennae were elongated. , The shape of stripe or even fragment, apoptotic bodies, DNA area was significantly reduced and other typical morphological characteristics of apoptotic cells. Conclusion Astaxanthin has a significant inhibitory effect on the proliferation of SGC-7901 cells and induces the apoptosis of gastric cancer cells.
