Objective To explore the influence of Toll-like receptor 2 (TLR2) on tumor angiogenesis and neoplasm development in B16 tumor bearing mice.Methods B16 tumor cells were injected into TLR2-/-mice to establish tumor bearing mice.In control group B16 tumor cells were injected into normal C57 mice.All groups were observed 4 weeks and tumor size was recorded.HSP60, IL-10 and TNF-αin serum were detected by ELISA method.TLR2,CD31, MMP-9 and MyD88 were masured by IHC and western blot.Results Compare the tumor growth curve between groups, in TLR2-/-the tumor volume decreased significantly.In TLR2 deficient mice, the level of HSP60 was decreased markedly compare to C57 tumor bearing mice.Expression of IL-10 and TNF-α in serum of the TLR2-/-mice were also significantly down regulated.In tumor tissues of TLR2-/-mice the expression of CD31, MMP-9 and MyD88 were obviously decreased.Conclusion Tumor angiogenesis and development were decreased in TLR2 knockout mice.TLR2 not only influences the pro-inflammation but also participates the tumor vessels formation and MMP-9 expression.TLR-2 might be a new potential target in melanoma treatment.
