1.RNA binding protein as target for antifibrotic therapy.Fibroproliferative disorders are characterized by excessive synthesis of type Ⅰ collagen.To find cure for fibrosis the molecular mechanism of collagen expression has to be elucidated.We discovered the specific regulation of type Ⅰ collagen expression,based on binding of RNA binding protein LARP6 to the stem-loop structure found in the 5 UTR of collagen mRNAs.Mutation of the 5 stem-loop or inactivation of LARP6 resulted in diminished collagen synthesis.LARP6 has a distinctive bipartite RNA binding domain and interacts only with collagen mRNAs with high affinity.Because 5 stem-loop is also unique for collagen rnRNAs,targeting the binding of LARP6 to 5 stem-loop can be specific and effective antifibrotic therapy.We have designed a cell based screening system for identification of chemical compounds that can displace LARP6 from 5 stem-loop.Use of this system will help discover a specific antifibrotic drug.
