Barley stripe mosaic virus (BSMV)-based virus-induced gene silencing (VIGS) is an effective and useful tool for rapid functional analysis of genes in wheat.The wheat phytoene desaturase (PDS) gene fragment was obtained by using RT-PCR from wheat.BSMV-γ vector and PDS fragment were digested by restriction enzymes and an recombinant BSMV-γ-TaPDS vector was constructed.One week after sap inoculation wheat plants,the infected plants showed obvious photobleaching symptom,while quantitative RT-PCR result demonstrated the decline ofPDS mRNA redundancy during infection.The established VIGS system will benefit the high-throughout function analysis of wheat genes in the following experiments.
