:
目的 构建人结肠癌裸鼠皮下移植瘤模型、经脾注射肝转移模型及盲肠原位移植瘤模型.方法 将人结肠癌细胞(HCT-116)接种于裸鼠皮下及脾被膜下,分别构建皮下移植瘤模型和经脾注射肝转移模型;以裸鼠皮下肿瘤为瘤源,将其剪切后移植于另一组裸鼠盲肠浆膜下,构建原位移植瘤模型.[结果]皮下移植瘤模型均有成瘤,但未见到局部侵袭、淋巴转移及血行转移等现象;经脾肝转移模型建模6 周后脾脏成瘤率为100%(6/6),

目的 构建人结肠癌裸鼠皮下移植瘤模型、经脾注射肝转移模型及盲肠原位移植瘤模型.方法 将人结肠癌细胞(HCT-116)接种于裸鼠皮下及脾被膜下,分别构建皮下移植瘤模型和经脾注射肝转移模型;以裸鼠皮下肿瘤为瘤源,将其剪切后移植于另一组裸鼠盲肠浆膜下,构建原位移植瘤模型.[结果]皮下移植瘤模型均有成瘤,但未见到局部侵袭、淋巴转移及血行转移等现象;经脾肝转移模型建模6 周后脾脏成瘤率为100%(6/6),肝转移率为66.7%(4/6),同时部分裸鼠出现了腹腔转移和腹水形成;盲肠原位移植瘤模型中,12 周后裸鼠盲肠原位成瘤率为80%(4/5),肝转移率为40%(2/5).[结论]皮下移植瘤模型可作为研究实体瘤局部生长的模型;经脾注射肝转移模型肝转移率较高,为研究结肠癌肝转移提供了可靠的模型;原位移植瘤模型最大程度地模拟了在体结肠癌的生物学行为,是较为理想的动物模型.
