Microevolution mechanism of probiotics Lactobacillus plantarum P-8 in human and rat intestinal tract

  Lactobacillus plantarum P-8 (Lp-8) is a novel probiotic bacterium isolated from naturally fermented yoghurt of Inner Mongolia.It has been reported that Lp-8 could modulate faecal bacterial structure,improve antioxidant capacity as well as lower blood lipid.However,we have few knowledge about mechanism of colonization and microevolution of Lp-8 in human or animal gut.In order to get insight into the mechanism,the current study,following a previous clinical study,detected the quantity of Lp-8 in faecal samples isolated before,during and after the comsumption by quantitative PCR,while reisolating the Lp-8 strains and sequencing whole genome of those strains.The result ofqPCR showed that after a daily oral dose of Lp-8 for 4 weeks,Lp-8 can be detected until the 17th week,while it couldnt be detected at the 4th week after a single intake only at day 0.Taking the origininal Lp-8 strain as reference,comparative genomics showed that a long period (17 weeks) colonization of Lp-8 could accumulate 9 SNPs,associated with different ages.And after a temprory (one day) intake,a short-term (2-3 weeks) stay in gut could result in 18 SNPs (17 were random mutation sites,1 was fixed).Moreover,no gain or loss of large fragment was detected among all the genomes.As a result,this study gives the first picture of adaptive microevolution of probiotics Lp-8 adapting to the complexity of the intestinal environment at genome level.
