At the upstream of yeast TORC1, a central regulatory factor of autophagy, there exists a hetero-dimeric G protein Gtr1/Gtr2 complex, which are homologues of mammalian Rag GTPases.We recently revealed that TORC1 is inactivated and autophagy is induced when Npr2/Npr3 inactivates Gtr1 and Gtr2 directely binds to TORC1 (Kira et al.Autophagy 2014).In mammals,Ragulator, which is consisted of p18/LAMTOR1, p14/LAMTOR2, MP1/LAMTOR3,HBXIP/LAMTOR4, C7orf59/LAMTOR5, functions as GEF for Rag GTPase.The possible counterpart of Ragulator in yeast is Ego complex, and it is consisted of Ego1 and Ego3 homodimers, but its exact composition still remains ambiguous.
