Analysis of Multilocus Sequence Typing of Listeria monocytogenes isolated from China

  [Objective]To understand the genetic diversity and distribution condition of Listeria monocytogenes strains isolated in China,we used the PasteurMLST database to obtain complete sequence types (ST) of L.monocytogenes strains from 1987 to 2015,and analyzed the clone complex and constructed the phylogenetic tree based on the nucleotide and amino acids sequence.[Methods]We used the SplitsTree 4.0 software to analyze the Split decomposition,the eBURST software to analyze the clone complexes and the MEGA 4.0 to obtain the phylogenetic tree of L.monocytogenes in China.[Results]The results showed that 53 items were obtained for analyzing the ST of Chinese L.monocytogenes strains from the database,which belonged to 44 ST types with ST705 type being the most dominant,and major of these strains were 1/2a serotype.All strains divided into two groups,which exited the genetic recombination.The eBURST software was used to analyze the data and three big clone complexes (CC155,CC1 and CC9) were observed from 1987 to 2015.The CC155 was the most prevalent.In addition,we obtained the CC9 and CC155 had closed evolutionary relationships by the analysis of phylogenetic in 7 housekeeping genes.Also,we found the genetic relationship among the isolates appears more closely on amino acids level than on nucleotide level.[Conclusion]Above all,Chinses L.monocytogenes strains are characterized by high polymorphism and can be subdivided into two groups.ST705 type is the most dominant strain,and CC155 was the most prevalent.It provided a theoretical foundation for further research on the origin and evolution ofL.monocytogenes.
