目的:建立用高效液相色谱法测定人血浆中卡铂浓度的方法。方法:采用Ultimate C18色谱柱(4.5mm×250mm,5μm);流动相为乙腈-0.02mol.L-1磷酸二氢钾溶液(0.5:99.5);流速1.0mL.min-1;检测波长230nm。结果:卡铂在0.77~49.07mg.L-1范围内线性关系良好(r=0.998),最低检测质量浓度为0.77mg.L-1,卡铂低、中、高浓度检测的日内、日间RSD分别为1.32%、4.25%、3.13%和2.77%、4.75%、3.30%。结论:该法具有操作简便可靠,准确,稳定性高等特点,适合应用于卡铂的治疗药物监测和药动学研究。
Objective: To establish a method for the determination of carboplatin concentration in human plasma by high performance liquid chromatography. Methods: Ultimate C18 column (4.5mm × 250mm, 5μm) was used. The mobile phase consisted of acetonitrile-0.02mol.L-1 potassium dihydrogen phosphate solution (0.5: 99.5), the flow rate was 1.0mL.min-1 and the detection wavelength was 230nm. Results: The linearity of carboplatin in the range of 0.77 ~ 49.07mg.L-1 was good (r = 0.998), the lowest detection concentration was 0.77mg.L-1. The intra- and inter-day RSD were 1.32%, 4.25%, 3.13% and 2.77%, 4.75% and 3.30% respectively. Conclusion: The method is simple, reliable, accurate and stable. It is suitable for the monitoring of therapeutic drugs and pharmacokinetics of carboplatin.
