Endoplasmic reticulum protein 29 (ERp29), one of the chaperones in endoplasmic reticulum (ER), essentially involves in protein unfolding and escort activity in the early secretary pathway.ER stress or other pathophysiological conditions will cause abnormal expression of ERp29.By dissecting the proteome profiling of Laser Capture Microdissected breast tumor cells, we identified that ERp29 is highly expressed in breast tumors, especially in HER-2/neu positive tumors.Overexpression of ERp29 in breast cancer cell line (MCF-7) and non-tumorigenic epithelial cell line (MCF12A) enhanced in vitro cell motility and invasion, colony-forming activity, and in vivo xenograft tumor growth.Our study indicates that ERp29 is a novel oncoprotein in breast tumorigenesis and a potential target in cancer treament.
