目的观察补肾和补肾健脾方对尾部悬吊骨丢失大鼠血清Ⅰ型胶原代谢的影响,比较二者防护骨丢失作用机制的异同。方法采用头低位-30°尾部悬吊模拟失重性骨丢失。大鼠随机分为正常、悬吊、补肾和补肾健脾共四组。实验结束后,双能X线骨密度分析仪检测右侧股骨骨密度,实时荧光定量PCR法检测左侧股骨骨组织Ⅰ型胶原的表达,ELISA法测血清PINP、PICP和ICTP含量。结果较之正常组,悬吊组大鼠股骨骨密度和骨组织Ⅰ型胶原的表达均显著降低(P<0.01),血清PINP、PICP和ICTP含量明显增高(P<0.01);较之悬吊组,补肾组和补肾健脾组大鼠股骨骨密度、骨组织Ⅰ型胶原的表达均升高(P<0.05,P<0.01),血清PINP、PICP和ICTP含量均降低(P<0.05,P<0.01);较之补肾组,补肾健脾组大鼠股骨骨密度和骨组织Ⅰ型胶原的表达均升高(P<0.05)、血清PINP、PICP和ICTP含量均降低(P<0.05,P<0.01)。结论补肾和补肾健脾方对模拟失重引起的废用性骨丢失具有防护作用,以补肾健脾方作用为优,其作用机制可能与改善Ⅰ型胶原代谢,进而增加骨组织Ⅰ型胶原表达有关。
Objective To observe the effects of Bushen and Bushen Jianpi Recipe on the serum type Ⅰ collagen metabolism in rats with tail suspension loss and to compare the mechanism of the two mechanisms in preventing bone loss. Methods The head low -30 ° tail suspension was used to simulate the loss of bone loss. Rats were randomly divided into normal, suspension, kidney and kidney invigorating the spleen in four groups. At the end of the experiment, the right femur BMD was detected by dual energy X-ray absorptiometry. The expression of collagen type Ⅰ in left femur bone was detected by real-time fluorescence quantitative PCR. The contents of PINP, PICP and ICTP in serum were detected by ELISA. Results Compared with normal group, the BMD of femur and the expression of type Ⅰ collagen in bone tissue of rats in suspension group were significantly decreased (P <0.01), and the contents of PINP, PICP and ICTP in serum were significantly increased (P <0.01) The BMD of femur and the expression of collagen type Ⅰ in bone, kidney and kidney invigorating spleen group were significantly increased (P <0.05, P <0.01), and the contents of PINP, PICP and ICTP in serum were lower (P <0.05, P <0.01). Compared with those in Bushen group, the BMD of femur and the expression of type Ⅰ collagen in bone-invigorating and invigorating kidney-invigorating group were significantly increased (P <0.05), and the levels of PINP, PICP and ICTP in serum were lower (P <0.05, P <0.01). Conclusion Bushen and Bushen Jianpi Recipe have a protective effect on bone loss due to simulated weightlessness, and the effect of Bushen Jianpi Fang is better. The mechanism may be related to the improvement of type Ⅰ collagen metabolism and the increase of type Ⅰ collagen expression in bone tissue .
