目的探讨Poly I:C对肠炎小鼠肠道黏膜IL-17的作用。方法 30只小鼠随机分为Poly I:C组、模型组、正常组,观察各组小鼠一般情况及结肠病理学变化,分光光度计测量结肠组织MDA、MPO的含量,免疫组织化学染色测定结肠组织中TNF-α、TGF-β、IL-17蛋白的表达。结果经Poly I:C治疗后,与模型组比较,小鼠一般情况及结肠病理均有明显改变,Poly I:C组MPO、MDA含量明显下降,模型组MPO、MDA含量明显升高,相比较差异有统计学意义(P<0.01),Poly I:C组结肠组织中TNF-α、IL-17、TGF-β蛋白的染色与模型组比较明显变浅。结论 Poly I:C可抑制肠炎结肠IL-17表达。
Objective To investigate the effect of Poly I: C on intestinal mucosal IL-17 in mice with enteritis. Methods Thirty mice were randomly divided into Poly I: C group, model group and normal group. The changes of colonic pathology and pathological changes in mice were observed. The contents of MDA and MPO in colonic tissues were measured by immunohistochemical staining The expression of TNF-α, TGF-β and IL-17 in colonic tissue. Results After treatment with Poly I: C, the general condition and pathological changes of the mice were significantly changed compared with the model group. The contents of MPO and MDA in Poly I: C group were significantly decreased, while the levels of MPO and MDA in model group were significantly increased The difference was statistically significant (P <0.01). The staining of TNF-α, IL-17 and TGF-βprotein in Poly I: C group was lighter than that in model group. Conclusion Poly I: C can inhibit the expression of IL-17 in colonic colon.
