目的探讨甲状腺乳头状癌(Papillary Thyroid Carcinomas,PTC)STAT3及其磷酸化水平与三叶因子3(Trefoil Factor 3,TFF3)表达及相关性。方法采用免疫组织化学检测31例组织芯片中PTC及癌旁组织STAT3、p STAT3和TFF3阳性率与平均光密度(AOD)值;Western Blot检测6例PTC和癌旁手术标本中相应蛋白的含量,并进行相关分析。结果 (1)免疫组化:TFF3蛋白在PTC中均高表达(P<0.001),TFF3阳性率与AOD值在直径>2 cm组、淋巴结转移组和临床Ⅲ~Ⅳ病例明显高于直径≤2 cm组、无淋巴结转移组和Ⅰ~Ⅱ期者(χ~2=12.120,P=0.001,χ~2=7.517,P=0.043,χ~2=10.490,P=0.003;AOD值:t=2.810,P=0.0064,t=2.234,P=0.039,t=2.926,P=0.0058);PTC中STAT3阳性率和AOD值与临床资料无关。p STAT3为胞核阳性,阳性率和AOD值在PTC组织低于癌旁组织,直径>2 cm组、淋巴结转移组和临床Ⅲ~Ⅳ病例明显低于直径≤2 cm组、无淋巴结转移组和Ⅰ~Ⅱ期者(χ~2=4.963,P=0.046,χ~2=4.882,P=0.033;AOD值:t=2.195,P=0.041,t=2.790,P=0.0068,t=2.018,P=0.042);TFF3与STAT3呈正相关,与p STAT3呈负相关(P<0.01)。(2)Western Blot:TFF3和STAT3蛋白在PTC的表达高于癌旁组织(t=2.841,P=0.008,t=2.925,P=0.007),但p STAT3却明显低于癌旁组织(t=2.713,P=0.009)。结论甲状腺乳头状癌TFF3和STAT3高表达、p STAT3低表达对PTC不良预后的预测有指导意义。
Objective To investigate the expression and correlation of STAT3, phosphorylation and Trefoil factor 3 (TFF3) in papillary thyroid papillary carcinoma (PTC). Methods Immunohistochemistry was used to detect the positive rate and average optical density (AOD) of STAT3, p STAT3 and TFF3 in PTC and paracancer tissues of 31 cases. Western Blot was used to detect the protein levels in 6 PTC and para- And make relevant analysis. Results (1) Immunohistochemistry: TFF3 protein was highly expressed in PTC (P <0.001). The positive rates of TFF3 and AOD were significantly higher in cases with diameter> 2 cm, lymph node metastasis and clinical cases Ⅲ ~ Ⅳ than those with diameters ≤2 cm group, no lymph node metastasis group and Ⅰ ~ Ⅱ phase (χ ~ 2 = 12.120, P = 0.001, χ ~ 2 = 7.517, P = 0.043, χ ~ 2 = 10.490, P = 0.003; , P = 0.0064, t = 2.234, P = 0.039, t = 2.926, P = 0.0058). The positive rate of STAT3 and AOD in PTC were not related to the clinical data. p STAT3 was positive for nucleus. The positive rate and AOD value were lower in PTC tissues than in paracancer tissues. The diameter> 2 cm group, lymph node metastasis group and clinical Ⅲ ~ Ⅳ cases were significantly lower than those of ≤2 cm in diameter and without lymph node metastasis AOD values: t = 2.195, P = 0.041, t = 2.790, P = 0.0068, t = 2.018, P = 0.046, χ ~ 2 = 4.882, P = = 0.042). TFF3 was positively correlated with STAT3 and negatively correlated with p STAT3 (P <0.01). (2) Western Blot: The expression of TFF3 and STAT3 in PTC was higher than that in paracancer tissues (t = 2.841, P = 0.008, t = 2.925, P = 0.007) 2.713, P = 0.009). Conclusion The high expression of TFF3 and STAT3 in papillary thyroid carcinoma and the low expression of p STAT3 may be helpful to predict the prognosis of PTC.
