视交叉上核(SCN)通过直接的纤维联系调控弓状核(ARC)的功能,ARC又通过调控穹窿下器(SFO)而调节心血管系统的功能。由于所采用方法的局限,对上述核团间的直接联系研究欠深入。为了进一步证实ARC与SFO神经元间以及SCN和ARC神经无间有无直接的纤维联系,本实验采用假狂犬病毒(PRV)跨神经元追踪、免疫荧光及免疫组化ABC法,通过病毒在各核团出现的时序来分析其间的联系。成年SD大鼠18只,体重25O-300克,动物行双侧颈上节切除术后,干单侧眼球内注射PRV3μ1,动物分别存活56、72、96小时。结果:①56h组,PRV阳性神经元主要出现在SCN腹外侧份,ABC法显示多数神经元系合VIP;②72h组,SCN内PRV阳性神经元增多,ARC内呈现出PRV阳性神经元,它们系小细胞神经元散在分布于整个核团。③96h组SFO内出现PRV阳性神经元,神经元体积较小,均匀散在分布。各时间组PRV阳性神经元脑体和树突均清晰可见。结果提示,动物存活56hSCN内出现PRV阳性神经元,经复制后,72h到达ARC,再经复制于96h到达SFO神经元。根据PRV阳性神经元出现的时序我们推测SCN与ARC神经元间以及ARC与SFO神经元间可能存在直接的纤维联系。SCN通过神经联系影响ARC的功能,ARC又通过直接的神经纤维联系参与SFO功能的调控。
The suprachiasmatic nucleus (SCN) regulates the function of the arcuate nucleus (ARC) through direct fiber connections, and ARC regulates the cardiovascular system by regulating the subfornular (SFO). Due to the limitation of the methods used, the research on the direct relationship between the above nuclei is underdeveloped. In order to further confirm the direct and indirect fiber connection between ARC and SFO neurons and between SCN and ARC nerves, we used pseudorabies virus (PRV) cross-neuron tracing, immunofluorescence and immunohistochemical ABC method to detect virus Troubles appear to analyze the connection between the time. Eighteen adult Sprague-Dawley rats weighing 25 to 300 grams were injected with PRV3μ1 in the unilaterally dry eye and the animals survived for 56,72 and 96 hours, respectively. Results: ① In group 56h, PRV-positive neurons mainly appeared in the ventral part of SCN. ABC method showed that most neurons were combined with VIP. ② In group 72h, PRV-positive neurons increased in SCN and PRV-positive neurons in ARC were smaller Cell neurons scattered throughout the nucleus. ③ PRV positive neurons were found in SFO of 96h group, neurons were smaller and distributed evenly. PRV positive neurons in each group of brain and dendrites are clearly visible. The results showed that PRV positive neurons appeared within 56hSCN of animals survived, and reached ARC at 72h after replication, and then reached SFO neurons at 96h after replication. According to the timing of occurrence of PRV-positive neurons, we speculate that there may be a direct fiber connection between SCN and ARC neurons and between ARC and SFO neurons. SCN affects the function of ARC through neural connection, and ARC also participates in the regulation of SFO function through direct nerve fiber connection.
