探讨淫羊藿甙逆转转化生长因子 β2 (TransformingGrowthFactorβ2 ,TGFβ2 )对LAK、CD3AK细胞的免疫抑制作用及其机制。方法 :采用MTT法 ,RT PCR和流式细胞术。结果 :淫羊藿甙能逆转受TGFβ2 抑制的LAK和CD3AK细胞的杀伤活性 ,并能部分恢复受TGFβ2 抑制的LAK细胞表面IL 2Rα表达和CD3AK细胞内穿孔素mRNA水平 ,以及CD3AK细胞的增殖活性。结论 :淫羊藿甙通过恢复LAK细胞IL 2Rα表达 ,提高CD3AK细胞内穿孔素mRNA水平及促进CD3AK增殖 ,逆转TGFβ2 对LAK、CD3AK的免疫抑制作用。
To investigate the effect of icariin on the immunosuppressive effects of Transforming Growth Foxβ (TGFβ2) on LAK and CD3AK cells and its mechanism. Methods: MTT assay, RT PCR and flow cytometry were used. Results: Icariin could reverse the cytotoxicity of LAK and CD3AK cells inhibited by TGFβ2 and partly restore the expression of IL-2Rα on LAK cells and the perforin mRNA levels in CD3AK cells inhibited by TGFβ2, as well as the proliferative activity of CD3AK cells. CONCLUSION: Icariin can reverse the inhibitory effect of TGFβ2 on LAK and CD3AK by restoring the expression of IL 2Rα in LAK cells, increasing the level of perforin mRNA in CD3AK cells and promoting the proliferation of CD3AK.
