目的:观察枸杞子含药血清对受损兔色素上皮细胞(RPE)中超氧化物歧化酶(SOD)活性及Bcl-2/Bax蛋白表达的影响。方法:常规细胞培养,建立兔RPE细胞光损伤模型,设正常对照组、模型组、低剂量组和高剂量组,分别于光照结束后24、48、72h进行指标的检测。结果:模型组各时间点兔RPE细胞中SOD活性及Bcl-2/Bax蛋白的表达与正常对照组比较,差异有统计学意义(P<0.05);低剂量组及高剂量组各时间点兔RPE细胞中SOD活性及Bcl-2/Bax蛋白表达与模型组比较,差异有统计学意义(P<0.05),且随时间的延长,SOD活性逐渐增高,Bcl-2蛋白表达逐渐增高,Bax蛋白表达依次降低,同时高、低剂量组之间存在量效关系。结论:枸杞子对兔RPE细胞具有一定的保护作用,这与其具有抗氧化性且能提高受损兔RPE细胞中SOD的活性有关。
Objective: To observe the effect of medlar serum on the activity of superoxide dismutase (SOD) and the protein expression of Bcl-2 / Bax in damaged rabbit pigment epithelial cells (RPE). Methods: The light injury model of rabbit RPE cells was established by routine cell culture. The normal control group, model group, low dose group and high dose group were established, and the indexes were detected at 24, 48 and 72 hours after light irradiation. Results: Compared with normal control group, the activity of SOD and the expression of Bcl-2 / Bax protein in RPE cells in model group at each time point had statistical significance (P <0.05). At each time point, the low dose group and high dose group Compared with model group, the activity of SOD and the expression of Bcl-2 / Bax in RPE cells were significantly different (P <0.05). With the prolongation of time, the activity of SOD and the expression of Bcl-2 protein gradually increased, The expression was decreased in turn, while there was a dose-response relationship between high and low dose groups. CONCLUSION: Fructus Lycii has a protective effect on RPE cells in rabbits, which is related to its antioxidant activity and the activity of SOD in RPE cells of injured rabbits.
