普鲁卡因酰胺(Ⅰ)的主要代谢产物N-乙酰普鲁卡因酰胺(Ⅱ)也有相似的抗心律失常效果。本文作者采用反相高压液相层析法进行血清中Ⅰ和Ⅱ的测定。层析柱3.9毫米(内径)×30厘米,填充剂:Bonda-pak C_(18),乙腈乙酸钠缓冲液洗脱,流速2毫升/分,压力2500psi,测定波长UV280毫微米。样品制备:在0.2毫升血清中加入0.3毫升水,0.1毫升Na_2CO_3缓冲液及4毫升乙醚,混合摇振30秒并离心,水层用干冰/丙酮冷冻,倾出有机层于室温中用N_2气流吹干,残余物加100微升乙腈乙酸钠缓冲液于室温中放置40分钟以保证Ⅰ
