目的探讨干扰素调节因子-5(IRF-5)rs2004640位点T等位基因与类风湿关节炎(RA)的临床表现的相关性。方法采用聚合酶链反应-连接酶检测反应(PCR-LDR)对136例RA患者进行IRF-5rs2004640位点的单核苷酸多态性(SNP)分型。记录患者的临床表现、X线分期,检测红细胞沉降率、类风湿因子水平。结果 X线分期为Ⅲ+Ⅳ期组RA患者的T等位基因阳性率显著高于Ⅰ+Ⅱ期组(P=0.018)。关节外表现<3个的RA患者的T等位基因阳性率显著多于关节外表现≥3个的RA患者(P<0.01)。结论 IRF-5 rs2004640 T等位基因与RA患者关节破坏程度(X线分期)和关节外表现有关。
Objective To investigate the relationship between the T allele of rs2004640 interferon regulatory factor-5 (IRF-5) and the clinical manifestations of rheumatoid arthritis (RA). Methods Single nucleotide polymorphisms (SNP) of IRF-5 rs2004640 were genotyped in 136 cases of RA patients by polymerase chain reaction-ligase assay (PCR-LDR). Record the clinical manifestations of patients, X-ray staging, detection of erythrocyte sedimentation rate, rheumatoid factor levels. Results The positive rate of T allele in patients with stage Ⅲ + Ⅳ RA was significantly higher than that in stage Ⅰ + Ⅱ (P = 0.018). RA patients with <3 extra-articular RA had significantly more positive T alleles than RA patients with ≥ 3 extra-articular manifestations (P <0.01). Conclusion The T allele of IRF-5 rs2004640 is associated with the degree of joint destruction (X-ray staging) and extra-articular manifestations in patients with RA.
