【目的】比较转移抑制基因nm2 3 H1表达产物核苷酸二磷酸激酶 (nucleosidediphosphatekinase ,NDPK)在转移力不同的肺癌克隆株中的表达。【方法】通过单细胞克隆技术及裸鼠接种筛选出高转移及低转移的人肺巨细胞癌克隆细胞株及高转移的人肺腺癌克隆细胞株 ,用LSAB免疫组化法检测nm2 3 H1基因产物NDPK的表达。【结果】NDPK在高转移及低转移的人肺巨细胞癌克隆细胞移植瘤及转移瘤中均有表达 ,高转移株的阳性细胞数目及强度均高于低转移株和裸鼠正常肺组织 ;NDPK在高转移的肺腺癌克隆细胞移植瘤中有表达 ,表达强度低于裸鼠正常肺组织。【结论】nm2 3 H1基因对肺巨细胞癌并不具转移抑制基因的功能 ,而可能促进肿瘤的发生、发展和转移。
【Objective】 To compare the expression of nm23 H1 mRNA in nucleus pulposus clones with different metastatic potential of nucleoside diphosphate kinase (NDPK). 【Method】 Human lung adenocarcinoma cell clones with high metastasis and low metastasis were screened by single cell cloning and nude mice inoculation, and cloned cell lines with highly metastatic human lung adenocarcinoma were detected by LSAB immunohistochemistry. Gene product NDPK expression. 【Results】 The expression of NDPK in both high-metastatic and low-metastatic human lung cancer cells and metastatic tumors was higher than that in the low-metastatic and normal lung tissues. NDPK was expressed in highly metastatic lung adenocarcinoma cell xenografts and its expression intensity was lower than that in normal lung tissue of nude mice. 【Conclusion】 The nm23 H1 gene does not function as a metastasis suppressor gene in lung giant cell carcinoma, but may promote tumorigenesis, development and metastasis.
