目的:探讨运动性疲劳后脊髓前角5-羟色胺(5-HT)及色胺酸羟化酶(TPH)表达的变化,阐明运动性疲劳在脊髓水平的发生机制。方法:健康成年雄性Wistar大鼠16只,随机分为运动疲劳组(E)和对照组(C),E组进行10天反复力竭运动,建立运动性疲劳动物模型。采用免疫组化方法和计算机图像分析技术,检测大鼠脊髓前角5-HT及TPH免疫反应阳性纤维及终末的平均积分光密度值。结果:E组大鼠脊髓前角5-HT及TPH阳性反应纤维和终末减少,平均积分光密度值显著低于C组(P<0.05)。结论:脊髓前角5-HT含量在运动性疲劳后降低提示脊髓前角5-HT与运动性疲劳的发生有关。而疲劳后前角TPH含量的降低可能是引起5-HT降低的关键因素。
Objective: To investigate the changes of serotonin (5-HT) and tryptophan hydroxylase (TPH) in the anterior horn of spinal cord after exercise-induced fatigue, and to elucidate the mechanism of exercise-induced fatigue in the spinal cord. Methods: Sixteen healthy male Wistar rats were randomly divided into exercise-induced fatigue group (E) and control group (C). E group was subjected to repeated exhaustive exercise for 10 days to establish a model of exercise-induced fatigue. Immunohistochemistry and computer image analysis were used to detect the mean integral optical density of 5-HT and TPH immunoreactive fibers in the anterior horn of spinal cord and the terminals. Results: The fibers and terminals of 5-HT and TPH-positive fibers in the anterior horn of spinal cord in group E were decreased, and the mean integrated optical density was significantly lower than that in group C (P <0.05). CONCLUSION: The decrease of 5-HT level in anterior horn of spinal cord after exercise-induced fatigue suggests that 5-HT of spinal cord anterior horn is associated with the occurrence of exercise-induced fatigue. The decrease of TPH content in the anterior horn after fatigue may be the key factor that causes the decrease of 5-HT.
