目的探讨丹酚酸B(SalB)对脑缺血高能磷酸化合物和乳酸含量的影响。方法复制急性脑缺血模型,测定缺血30 m in脑组织高能磷酸化合物和乳酸的含量以及脑组织的含水量。结果缺血组脑组织ATP[(12.12±3.71)μmol/g]和磷酸肌酸[(65.24±8.84)μmol/g]含量及Na+-K+-ATPase活性[(1.321±0.373)U/mg]显著降低(P<0.01),而乳酸[(3.58±0.81)mmol/g]含量显著升高(P<0.01)、脑指数[(1.508±0.002)%]和脑含水量[(78.75±0.47)%]明显增多(P<0.05~0.01)。用丹酚酸B治疗,脑组织ATP[(18.16±3.38)μmol/g]和磷酸肌酸[(74.46±6.58)μmol/g]含量显著升高(P<0.01),Na+-K+-ATPase[(1.902±0.491)U/mg]活性增强(P<0.05),而乳酸[(2.59±0.41)mmol/g]含量明显减少(P<0.01)、脑指数[(1.505±0.002)%]和脑含水量[(78.10±0.44)%]则降低(P<0.05)。结论增加高能磷酸化合物以及降低乳酸的含量,可能是丹酚酸B减轻急性脑缺血早期脑水肿的作用机制。
Objective To investigate the effects of salvianolic acid B (SalB) on cerebral ischemia-induced high-energy phosphate compounds and lactic acid content. Methods The acute cerebral ischemia model was established. The content of high-energy phosphate compounds and lactic acid in brain tissue and the water content in brain tissue were determined 30 min after ischemia. Results The contents of ATP [(12.12±3.71) μmol/g] and phosphocreatine [(65.24±8.84) μmol/g] and Na+-K+-ATPase activity [(1.321±0.373) U/mg] were significant in the ischemic brain tissue. Decreased (P<0.01), while lactic acid [(3.58±0.81) mmol/g] content increased significantly (P<0.01), brain index [(1.508±0.002)%] and brain water content [(78.75±0.47)% ] Significantly increased (P<0.05~0.01). Treatment with salvianolic acid B significantly increased brain ATP [(18.16±3.38) μmol/g] and creatine phosphate [(74.46±6.58) μmol/g] (P<0.01), and Na+-K+-ATPase [ (1.902±0.491) U/mg] activity increased (P<0.05), while lactate [(2.59±0.41) mmol/g] content decreased significantly (P<0.01), brain index [(1.505±0.002)%] and brain Water content decreased ((78.10±0.44)%) (P<0.05). Conclusion Increasing the amount of high-energy phosphate compounds and decreasing the content of lactic acid may be the mechanism by which salvianolic acid B can reduce cerebral edema in the early stage of acute cerebral ischemia.
