目的观察亚甲蓝对百草枯诱导急性肺损伤的保护作用。方法取清洁级雄性SD大鼠24只,随机分为空白对照组、百草枯组、亚甲蓝组和百草枯+亚甲蓝组,每组6只。腹腔一次性注射25 mg/kg百草枯建立大鼠急性中毒模型,亚甲蓝治疗组大鼠在百草枯给药2 h后给予2 mg/kg亚甲蓝。24 h后麻醉、固定,取肺组织,测量肺湿/干重比及观察肺组织病理学变化;取肺组织匀浆,检测超氧化物歧化酶(SOD)和乳酸脱氢酶(LDH)活性及丙二醛(MDA)含量;提取肺泡灌洗液检测髓过氧化物酶(MPO)活性。结果与空白对照组及亚甲蓝组相比,百草枯组大鼠肺组织湿/干重比增加(P<0.05),大鼠肺组织病理学评分增加(P<0.05)、肺组织SOD活性降低(P<0.05)、MDA和LDH含量增加(P<0.05)、肺泡灌洗液中MPO活性增加;与百草枯组相比,百草枯+亚甲蓝组大鼠肺湿/干重比降低(P<0.05),肺组织病理学评分降低(P<0.05),肺组织SOD活性增高(P<0.05),MDA和LDH含量减少(P<0.05)且肺泡灌洗液中MPO活性降低(P<0.05)。结论亚甲蓝通过抗氧化作用对百草枯诱导的大鼠急性肺损伤发挥保护作用。
Objective To observe the protective effect of methylene blue on paraquat-induced acute lung injury. Methods Twenty-four male SD rats were randomly divided into blank control group, paraquat group, methylene blue group and paraquat + methylene blue group, with 6 rats in each group. Acute poisoning model was established by intraperitoneal injection of 25 mg / kg paraquat. The rats in methylene blue treatment group were given 2 mg / kg methylene blue 2 h after paraquat administration. Twenty-four hours later, the rats were anesthetized, fixed, and lung tissue was taken out. The wet / dry weight ratio of lungs and the histopathological changes of lung were observed. The lung homogenates were taken for measuring the activities of superoxide dismutase (SOD) and lactate dehydrogenase (LDH) And malondialdehyde (MDA) were measured. The activity of myeloperoxidase (MPO) was detected by the extraction of BALF. Results Compared with the blank control group and the methylene blue group, the lung wet / dry weight ratio in the paraquat group increased (P <0.05), the lung pathological score increased (P <0.05), the SOD activity in the lung tissue (P <0.05), the content of MDA and LDH increased (P <0.05), and the activity of MPO in BALF increased. Compared with paraquat group, the lung wet / dry weight ratio of paraquat + methylene blue group decreased (P <0.05). The lung histopathological score decreased (P <0.05), the activity of SOD in lung tissue increased (P <0.05), MDA and LDH decreased (P <0.05), and the activity of MPO in bronchoalveolar lavage fluid decreased <0.05). Conclusion Methylene blue can protect paraquat-induced acute lung injury in rats through antioxidation.
