背景:以往多应用大鼠建立肝移植模型对肝移植术后排斥反应进行基础研究,而灵长类非人类大动物建立肝移植模型能更接近临床肝移植的需求。目的:探讨建立稳定的恒河猴原位肝移植模型的最佳方案。方法:选用健康恒河猴进行同种异体原位肝移植20次,以10只雄性作为受体,10只雌、雄不限为供体,借鉴临床肝移植和各种动物模型建立的方法,使用二袖套法加肝动脉重建建立稳定的恒河猴原位肝移植模型。结果与结论:20次恒河猴原位肝移植模型成功率90%,供肝切取时间(17±3)min,供肝修整时间(35±5)min,受体移植时间(133±45)min,受体无肝期(12±4)min,术后24h存活率90%(18/20),72h存活率为80%(16/20),1周存活率50%(10/20),14只分别于术后2周内死于急性排斥反应,最长存活38d,也死于急性排斥反应,所有受体均无门静脉血栓形成及胆道并发症发生。改进后的恒河猴同种异体原位肝移植模型具有操作简便、手术成功率高、重复性和稳定性好的优点,是肝移植临床前期研究的较理想动物模型。
BACKGROUND: In the past, multi-application rat models of liver transplantation were established to conduct basic researches on rejection after liver transplantation. However, the establishment of liver transplantation model by primate non-human animals can be closer to the needs of clinical liver transplantation. Objective: To explore the best way to establish a stable rhesus monkey orthotopic liver transplantation model. Methods: Healthy rhesus monkeys were used for allogeneic orthotopic liver transplantation 20 times. 10 males were used as recipients and 10 males and females as donors. Based on the clinical liver transplantation and various animal models, Establishment of a stable rhesus monkey orthotopic liver transplantation model by two cuffs plus hepatic artery reconstruction. RESULTS AND CONCLUSION: The success rate of orthotopic liver transplantation in 20 rhesus macaques was 90%, the time of hepatectomy (17 ± 3) min, the time of donor liver dressing (35 ± 5) min, the time of recipient transplantation (133 ± 45) The survival rate was 90% (18/20) at 24 hours after operation, 80% (16/20) at 72 hours and 50% at 1 week (10/20) , 14 died of acute rejection within 2 weeks after operation respectively. The longest survival was 38 days and died of acute rejection. No portal vein thrombosis and biliary complications occurred in all the recipients. The improved rhesus monkey heterotopic orthotopic liver transplantation model has the advantages of simple operation, high success rate, good repeatability and good stability, and is an ideal animal model for preclinical liver transplantation.
