目的观察脂肪干细胞(ADSCs)源性神经干细胞克隆球的超微结构形态。方法原代培养大鼠ADSCs,向神经干细胞诱导分化,分化为克隆球后,免疫荧光鉴定其Nestin表达,透射电子显微镜观察克隆球超微结构形态。结果大鼠ADSCs可诱导分化为细胞克隆球,其Nestin表达阳性,超微结构与原代培养的神经干细胞克隆球的超微结构相似。结论 ADSCs经诱导分化后形成的克隆球具有神经干细胞的特征。
Objective To observe the ultrastructural morphology of the cloned spheres of adipose derived stem cells (ADSCs)-derived neural stem cells. Methods Primary cultured rat ADSCs were induced to differentiate into neural stem cells, and then differentiated into cloned spheres. Nestin expression was identified by immunofluorescence. The ultrastructure of the cloned spheres was observed by transmission electron microscopy. Results Rat ADSCs could be induced to differentiate into spheres. The expression of Nestin was positive. The ultrastructure was similar to the ultrastructure of primary cultured NSCs. Conclusion The cloned spheres formed after differentiation of ADSCs have the characteristics of neural stem cells.
