对影响小麦幼胚愈伤组织高频率再分化的条件进行了研究。结果表明,暗培养条件下诱导的小麦幼胚愈伤组织的再分化频率显著高于散光和2000lx光照条件;大田生长的小麦比温室盆栽小麦的幼胚脱分化和再分化频率高。小麦主茎穗部的幼胚组织脱分化和再分化特性显著高于小麦分蘖茎穗部的幼胚组织。不同年份、不同基因型小麦品种,及不同胚龄幼胚组织的脱分化和再分化特性有一定差异。通过诸多因素的优化和最适幼胚组织的选择,可建立小麦基因转化中不依赖于基因型的良好受体系统。
The conditions affecting the high frequency of redifferentiation of wheat immature embryos were studied. The results showed that the frequency of redifferentiation of wheat immature embryos induced by dark culture was significantly higher than that of astigmatism and 2000lx light conditions. The frequency of dedifferentiation and redifferentiation of young wheat grown in field was higher than that of greenhouses. The immature embryo dedifferentiation and re-differentiation characteristics of the main stem of wheat were significantly higher than that of the young embryo of the tillering stem of wheat. There were some differences in dedifferentiation and redifferentiation characteristics between different wheat genotypes and different embryo age immature embryos. Through the optimization of many factors and the selection of the most suitable immature embryo tissue, a good receptor independent genotype-free system can be established in wheat gene transformation.
