目的:研究合成冰片对非孕大鼠离体子宫平滑肌运动的影响及其作用机制,为临床安全合理使用冰片提供参考。方法:采用BL-420生物机能实验系统,观察不同浓度冰片对非孕大鼠离体子宫平滑肌的影响;并观测一定浓度冰片对缩宫素、氯化乙酰胆碱、阿托品等工具药对子宫平滑肌的部分影响机制。结果:①冰片对非孕大鼠离体子宫平滑肌呈先兴奋后抑制的作用,且抑制作用随浓度递增而增强,当冰片的终浓度为3.92×10-2mg/ml时,子宫平滑肌呈现明显的松弛状态。②冰片2.63×10-2mg/ml能明显对抗缩宫素引起的收缩张力、峰面积、频率增加(P<0.01);再次加入缩宫素能明显增强冰片后的子宫平滑肌的收缩频率(P<0.01)。③冰片2.63×10-2mg/ml能显著对抗氯化乙酰胆碱引起的收缩张力、峰面积、频率增加(P<0.01),再次加入氯化乙酰胆碱,对冰片后的子宫平滑肌没有明显影响;冰片2.63×10-2mg/ml能协同阿托品进一步抑制子宫平滑肌的张力、峰面积(P<0.05)、频率(P<0.01),加入氯化乙酰胆碱,对阿托品与冰片处置后的离体子宫平滑肌未能产生显著性兴奋作用。结论:冰片对非孕大鼠离体子宫平滑呈现显著抑制作用,并能抑制缩宫素、乙酰胆碱所致的子宫平滑肌兴奋,有拟阿托品样作用,推测冰片通过部分阻断抗利尿激素受体和完全阻断M受体而发挥抑制效应。
Objective: To study the effect of synthetic borneol on uterine smooth muscle movement in non-pregnant rats and its mechanism of action, and provide a reference for safe and rational use of borneol. Methods: BL-420 bio-functional experimental system was used to observe the effects of different concentrations of borneol on isolated uterine smooth muscle in non-pregnant rats. The effects of some borneol on oxytocin, acetylcholine, atropine, Influence Mechanism. RESULTS: (1) The borneol had an inhibitory effect on isolated uterine smooth muscle of non-pregnant rats, and the inhibitory effect increased with increasing concentration. When the final concentration of borneol was 3.92 × 10-2mg / ml, the uterine smooth muscle showed obvious Relaxed state. ② Borneol 2.63 × 10-2mg / ml obviously antagonizes the contraction tension, peak area and frequency caused by oxytocin (P <0.01). The addition of oxytocin significantly enhances the contractile frequency of uterine smooth muscle after borneol treatment (P < 0.01). ③ Borneol 2.63 × 10-2mg / ml can significantly antagonize the acetylcholine chloride induced contraction tension, peak area, frequency increased (P <0.01), re-add acetylcholine, borneol uterine smooth muscle had no significant effect; borneol 2.63 × 10-2mg / ml synergistic atropine can further inhibit uterine smooth muscle tension, peak area (P <0.05), frequency (P <0.01), adding acetyl chloride, atropine and borneol treatment of isolated uterine smooth muscle failed to produce significant Sexual excitement. CONCLUSION: Borneol significantly inhibits uterine smoothness in isolated non-pregnant rats and inhibits the excitement of uterine smooth muscle cells induced by oxytocin and acetylcholine. Simultaneously, borneol partially blocks the anti-vasopressin receptor and M block completely blocked and exert inhibitory effect.
