目的:优选菟丝子的酶法提取工艺,以提高菟丝子中金丝桃苷及总黄酮的提取率。方法:以金丝桃苷提取量为指标,通过单因素试验考察酶种类、酶用量、酶解时间、酶解p H及温度对酶解效果的影响;以金丝桃苷与总黄酮提取量的综合评分为指标,采用正交试验考察酶用量、酶解p H及酶解温度对酶解工艺的影响。采用HPLC测定金丝桃苷含量,流动相乙腈-0.1%磷酸溶液(17∶83),检测波长360 nm。结果:选择纤维素酶,最佳提取工艺为酶用量2.0%,酶解时间2.0 h,酶解p H 4.0,酶解温度45℃;金丝桃苷、总黄酮提取量分别为1.408,26.966 mg·g-1。结论:优选的酶法提取工艺稳定合理,适用于菟丝子的工业生产。
Objective: To optimize the enzymatic extraction of dodder to improve the extraction rate of hyperoside and total flavonoids in dodder. Methods: According to the amount of hyperoside extracted, the effects of enzyme type, enzyme dosage, enzymolysis time, enzymatic hydrolysis p H and temperature on the enzymatic hydrolysis were investigated by single factor test. The content of hyperoside and total flavonoids The comprehensive score as an indicator, using orthogonal test to examine the amount of enzyme, enzymatic hydrolysis p H and enzymatic hydrolysis temperature on the enzymatic hydrolysis process. The content of hyperoside was determined by HPLC. The mobile phase was acetonitrile-0.1% phosphoric acid solution (17:83). The detection wavelength was 360 nm. Results: The best extraction technology was 2.0% enzyme dosage, 2.0 h hydrolysis time, p H 4.0 enzymolysis temperature and 45 ℃ enzymolysis temperature. The content of hyperoside and total flavonoids were 1.408 and 26.966 mg · G-1. Conclusion: The optimal enzymatic extraction process is stable and reasonable, suitable for industrial production of dodder.
