目的:探讨雷公藤多苷(glycosides of Tripterygium wilfordii,GTW)对雄性幼鼠生殖损伤和损伤后最终生育能力的影响及补肾中药(菟丝子黄酮、六味地黄丸)的保护作用。方法:①生殖损伤研究:3~4周龄SD雄性幼鼠48只,随机分为4组:空白组(CMC-Na)、雷公藤多苷组(9 mg·kg-1·d-1)单独ig,六味组(六味地黄丸1 g·kg-1·d-1+GTW 9 mg·kg-1·d-1),黄酮组(菟丝子黄酮0.1 g·kg-1·d-1+GTW 9 mg·kg-1·d-1)混合ig,持续12周后,处死取一侧睾丸、附睾立即称质量,计算脏器指数,称重后放入固定液中保存,待观察睾丸组织形态学改变;②生育能力研究:成年大鼠100只,每组25只,分组喂养方法同上,持续12周后,将所有大鼠分笼喂养,每笼1只,按照1∶1比例放入成年雌性大鼠1只,分3次合笼,每次合笼时间为15 d,合笼后观察雌鼠受孕率、仔鼠离乳存活率、仔鼠生长发育情况。结果:雷公藤多苷对睾丸病理组织生精细胞肿胀变性,精原细胞、精母细胞、精子细胞明显减少,菟丝子黄酮、六味地黄丸能明显减轻这种损伤;用药12周后雄性幼鼠体重、睾丸质量、睾丸指数、附睾质量、附睾指数与空白组相比无显著性差异(P>0.05);每次合笼后受孕率、离乳存活率、幼鼠的生长发育与空白组相比无显著性差异。结论:雷公藤多苷对雄性幼鼠生殖系统的损伤是可逆的,对睾丸的病理组织损伤是可以恢复的,补肾中药具有保护这种病理组织损伤作用;雷公藤多苷对最终的生育能力没有影响,对幼仔生长发育没有影响。

Results: Tripterygium glycosides on spermatogenic cells of the testicular pathological changes of swelling and degeneration, spermatogonia, spermatocytes, sperm cells decreased significantly, Dusanzi, Liu Wei Di Huang Wan can significantly reduce this damage; 12 weeks after treatment male rats weight , Testicular mass, testicular index, epididymis quality and epididymis index had no significant difference compared with the blank group (P> 0.05). The pregnancy rate, the lactation survival rate, the growth and development of the young rats No significant difference. CONCLUSION: Tripterygium glycosides can damage the reproductive system of male pups, and can restrain pathological injury of testis. TCM of tonifying kidney can protect the pathological tissues from injury. Tripterygium glycosides have no effect on the final fertility Influence, no effect on the growth and development of pups.
