为了解白血病干细胞 (stemcellleukemia ,SCL)基因在白血病骨髓基质细胞 (BMSC)及骨髓细胞中的表达情况 ,收集 18例急性髓系白血病 (AML)、17例慢性粒细胞白血病 (CML)、7例急性淋巴细胞白血病 (ALL)和 33例正常骨髓标本的单个核细胞 (MNC)进行体外长期培养 ,分别收集悬浮细胞 (造血细胞 )和扩增后的贴壁细胞 (BMSC)。运用RT PCR ELISA检测SCL基因的表达 ,分析表达率 ,并以管家基因 β2 微球蛋白 (β2 M)为内参照进行半定量分析。结果发现 ,SCL基因在AML(2 7.8% )和CML(11.8% )的BMSC中的表达率均低于正常对照组 (6 9 7% ,P <0 .0 5 )。SCL基因在CML骨髓造血细胞中的表达率 (6 4 .3% )高于其对应的BMSC(P <0 .0 5 )。半定量分析SCL基因在AML骨髓造血细胞中的表达水平显著高于其对应的BMSC(P <0 .0 5 )。结论 :SCL基因在AML和CML的BMSC中的相对低表达状态可能与血液病造血调控的异常有关。
18 acute myeloid leukemia (AML), 17 chronic myeloid leukemia (CML), and 7 acute myeloid leukemia (AML) were collected to investigate the expression of leukemic stem cell (SCL) gene in leukemia bone marrow stromal cells (BMSC) Lymphocytic leukemia (ALL) and 33 normal bone marrow samples of mononuclear cells (MNC) were cultured in vitro for long-term culture. Resuspended cells (hematopoietic cells) and expanded adherent cells (BMSCs) were collected. The expression of SCL gene was detected by RT PCR ELISA. The expression of SCL gene was analyzed by semi-quantitative analysis with housekeeping gene β2 microglobulin (β2 M) as an internal reference. The results showed that the expression rates of SCL gene in BMSCs of AML (2 7.8%) and CML (11.8%) were lower than that of the normal control group (69.7%, P <0.05). The expression rate of SCL gene in CML bone marrow hematopoietic cells (64.3%) was higher than that of corresponding BMSC (P <0.05). Semi-quantitative analysis of SCL gene expression in AML bone marrow hematopoietic cells was significantly higher than its corresponding BMSC (P <0.05). Conclusion: The relative low expression of SCL gene in BMSCs of AML and CML may be related to abnormal regulation of hematopoietic diseases.
