目的研究肺癌细胞DNA倍体类型和核仁组织区嗜银蛋白(AgNOR)及两者之间的相互关系。方法对36例肺癌石蜡标本进行细胞AgNOR定量计数和DNA含量的流式细胞术(FCM)检测。检测结果进行t检验和X2检验。结果二倍体肺癌细胞AgNOR计数(1.65±0.87)显著低于异倍体肺癌细胞者(3.09±1.03)细胞周期中S期细胞<20%的肺癌细胞AgNOR计数(1.38±1.02)显著低于S期细胞≥20%者(2.77±1.14)。在二倍体肿瘤中,分化Ⅱ级和Ⅲ级细胞的AgNOR计数显著高于分化Ⅰ级者,但在Ⅱ、Ⅲ级之间AgNOR计数则无显著差异;分化Ⅲ级肿瘤的异倍体检出率显著高于分化Ⅰ、Ⅱ级肿瘤,但在Ⅰ、Ⅱ级之间异倍体检出率则无显著差异。结论细胞DNA含量和AgNOR计数与肺癌细胞的增殖状态、恶性程度及预后有密切相关性;在病理分级中以上两种检测具有互补性。
Objective To study the relationship between the DNA ploidy type and AgNOR and their relationship in lung cancer cells. Methods Quantitative cell counts and flow cytometry (FCM) were performed on 36 paraffin-embedded lung cancer specimens. The test results were t-test and X2 test. Results AgNOR counts of diploid lung cancer cells (1.65±0.87) were significantly lower than those of aneuploid lung cancer cells (3.09±1.03). 1.38±1.02) was significantly lower than ≥20% of S-phase cells (2.77±1.14). In diploid tumors, AgNOR counts of differentiated grade II and grade III cells were significantly higher than those of grade I differentiation, but there was no significant difference in AgNOR counts between grades II and III; the heteroploid detection rate of differentiated grade III tumors Significantly higher than the differentiation of grade I and II tumors, but there was no significant difference in the detection rate of heteroploid between grades I and II. Conclusions Cellular DNA content and AgNOR counts are closely related to the proliferation status, malignancy and prognosis of lung cancer cells. The above two detection methods are complementary in pathological grade.
