胃癌细胞中RHAMM基因表达的对比分析

目的 分析胃癌发生中透明质酸 (hyaluronic acid,HA)介导的细胞游走受体 (receptor of HAmediated motility,RHAMM)的表达形式以及 HA对其表达的影响。方法 采用免疫组织化学和 Western印迹杂交方法 ,分析 RHAMM蛋白在 10例萎缩性胃炎、12例肠上皮化生、2 0例 度和 15例 度非典型增生、74例各类型胃癌 / 2株胃癌细胞系以及 36例非癌粘膜临床标本中的表达形式。结果  17%的非癌粘膜有相对分子质量 (Mr)为 95 0 0 0和 93 0 0 0的 RHAMM低水平表达。RHAMM阳性率随癌前病变的进展而显著提高 ,在高、中、低和未分化癌中分别达 70 %、82 %、71%和 6 8%。70 %以上的 RHAMM阳性胃癌病例 ,除 Mr93 0 0 0~ 95 0 0 0胞内蛋白外 ,还可见诸如 Mr73 0 0 0、70 0 0 0和 6 0 0 0 0等多种拼接异构物。常规培养的胃癌细胞仅表达胞内型 RHAMM(IHABP)。HA可明显促进 RHAMM拼接异构 ,使被处理细胞出现胞膜阳染。结论 RHAMM基因在非癌和胃癌组织中的表达和存在方式不同 (P<0 .0 1) ;膜型 RHAMM蛋白的出现与胃粘膜恶性转化呈正相关 (P<0 .0 1) ;HA对 RHAMM的拼接异构表达具有调节作用 Objective To analyze the expression of hyaluronic acid (HA)-mediated receptor of HA mediated motility (RHAMM) and the effect of HA on the expression of gastric cancer. Methods Immunohistochemistry and Western blot were used to analyze RHAMM protein in 10 cases of atrophic gastritis, 12 cases of intestinal metaplasia, 20 cases and 15 cases of atypical hyperplasia, and 74 cases of various types of gastric cancer/2 gastric cancer cells. Department and 36 cases of non-cancerous mucosa in clinical specimens. Results 17% of non-cancer mucosa had low molecular weight (Mr) of 9500000 and 930000 low-level expression of RHAMM. The positive rate of RHAMM increased significantly with the progression of precancerous lesions, reaching 70%, 82%, 71%, and 68% in high, middle, low, and undifferentiated cancers, respectively. More than 70% of RHAMM positive gastric cancer cases, in addition to Mr930000 ~ 9500000 intracellular protein, also can be seen such as Mr70000, 70000 and 600000 and other splicing isomers. Conventionally cultured gastric cancer cells only express intracellular RHAMM (IHABP). HA can significantly promote RHAMM splicing isomerization, so that the treated cells appear membrane staining. Conclusions RHAMM gene expression is different in the non-cancer and gastric cancer tissues (P<0.01).
