人体液中胆汁酸水平异常能灵敏地反映出消化系统病变。胆汁酸测定对于肝、胆病变诊断、治疗及基础医学研究有重要价值。 本法用反相色谱分离胆汁酸,柱后串接3α-羟基类固醇脱氢酶(3α-HSD)固相酶柱,胆汁酸在酶催化下,将底物辅酶Ⅰ(NAD)定量转换成NADH,荧光检测NADH,从而定量测定胆汁酸。方法特异性强,灵敏度高,样品处理简便,分析速度快。该法国外已有报道,国内尚未见。
Abnormal human bile acid levels can sensitively reflect the digestive system lesions. Bile acid determination for liver and gallbladder disease diagnosis, treatment and basic medical research has important value. In this method, bile acid is separated by reverse phase chromatography, and the 3α-hydroxysteroid dehydrogenase (3α-HSD) immobilized enzyme column is connected to the column. The bile acid is catalytically converted to NADH , Fluorescent detection of NADH, which quantitative determination of bile acids. Method specificity, high sensitivity, sample processing is simple, fast analysis. It has been reported outside France that it has not been seen in the country yet.
