目的 研究白细胞介素— 1 β(IL 1 β)在大鼠脑缺血后的表达情况 ,以探讨脑缺血后脑内是否有内源性的IL 1β产生 ,以及由何种细胞产生。方法 采用线堵法使右侧大脑中动脉永久堵塞制成大鼠局灶脑缺血模型 ,用免疫组化方法 ,显示IL 1β在脑内的表达。结果 正常和假手术大鼠脑内以及脑缺血后的非缺血半球未发现有IL 1β免疫反应阳性细胞。脑缺血 3小时后 ,在缺血区有散在的IL 1 β阳性细胞 ,呈未活化的星形胶质细胞形态 ,随缺血时间延长阳性星形胶质细胞数增多 ,且活化程度越高 ,到 2个月时 ,坏死灶周围仍有许多IL 1 β阳性、明显活化的星形胶质细胞。结论 脑缺血后3小时至 2个月 ,脑内有内源性的、蛋白水平的IL 1β产生 ,来源为星形胶质细胞。IL 1β可能与星形胶质细胞增生及疤痕形成有密切的关系。
Objective To investigate the expression of interleukin - 1β (IL - 1β) after cerebral ischemia in rats and to explore whether endogenous IL - 1β is produced in the brain and which cells produce it. Methods The occlusion of right middle cerebral artery was blocked by the method of line blockage to establish the model of focal cerebral ischemia in rats. Immunohistochemistry was used to show the expression of IL-1β in the brain. Results No IL 1β immunoreactive cells were found in normal and sham-operated rats and in non-ischemic hemisphere after cerebral ischemia. Three hours after cerebral ischemia, interspersed IL 1 β positive cells in the ischemic area showed non-activated astrocyte morphology. The number of positive astrocytes increased and the degree of activation increased with prolonged ischemia By 2 months, there were still many IL-1β-positive and markedly activated astrocytes around the necrotic area. Conclusions From 3 hours to 2 months after cerebral ischemia, there is an endogenous and protein level of IL-1β in the brain, which is derived from astrocytes. IL 1β may be closely related to astrogliosis and scarring.
