建立一种简捷、灵敏和直观的方法,测定木聚糖在实验大鼠肠道各个区段消化率。将20只健康SPF级SD雄性大鼠按体重随机分成2组,每组10只,分别喂以含0%、3%木聚糖的高脂高糖饲料7周后,将含3%木聚糖的饲料更换为含3%不溶性气巴蓝着色木聚糖的饲料,饲喂1周后颈椎脱臼法处死大鼠,快速解剖,收集空肠、回肠、盲肠、结肠和直肠内容物,根据浸提液625 nm处的吸光值得出气巴蓝染料含量,并换算出木聚糖的肠道消化率。结果表明,木聚糖到达空肠、回肠、盲肠、结肠段的消化率分别为0%、6.9%、62.5%和85.5%,到直肠段的消化率可达100%,人工提取的木聚糖可在大鼠肠道内被微生物充分降解,并基本均匀地分布于整个大肠段内消化,且可输送到最远端的直肠段才会被消化完全。我们的结果表明木聚糖是一种优质膳食纤维资源。本文建立的不溶性显色基质法测定木聚糖在肠道中的消化率,具有简捷、灵敏和直观的效果,并为其它膳食纤维的消化特性研究提供了有益的启示。
A simple, sensitive and intuitive method was established to determine the digestibility of xylan in various sections of intestine in experimental rats. Twenty healthy SPF SD male rats were randomly divided into two groups according to body weight. Ten rats in each group were fed with high-fat and high-sugar diet containing 0% and 3% xylans for 7 weeks. The swine feed was changed to 3% insoluble gas-barn-colored xylan feed, rats were sacrificed by cervical dislocation after 1 week of feeding, dissected quickly, and jejunum, ileum, cecum, colon, and rectal contents were collected, Absorbance at 625 nm gives the content of gas-blue dye and translates into intestinal digestibility of xylan. The results showed that the digestibility of xylan to the jejunum, ileum, cecum and colon was 0%, 6.9%, 62.5% and 85.5%, respectively. The digestibility of xylan to the rectum was up to 100% In the intestine of rats, it is fully degraded by microorganisms, and is uniformly distributed throughout the large intestine and digested, and can be delivered to the distal rectal segment to be completely digested. Our results show that xylan is a good source of dietary fiber. The insoluble chromogenic matrix method established in this paper was used to determine the digestibility of xylan in the intestine. The method has the advantages of simplicity, sensitivity and visualization, and provides useful enlightenment for the digestive characterization of other dietary fiber.
