建立海洛因成瘾动物模型,研究海洛因成瘾大鼠血清代谢物变化,寻找其潜在的标志物。实验动物设成瘾组和空白对照组,运用液相色谱联合离子阱-飞行时间质谱(LC/MS-IT-TOF)分析方法研究海洛因成瘾对大鼠血清代谢变化的影响。分别采用主成分分析(PCA)、偏最小二乘法-判别分析(PLS-DA)分析成瘾组和正常对照组之间的代谢差异。结果表明成瘾组和对照组大鼠在血清代谢水平上具有明显差异,经统计学分析初步筛选出26个潜在的生物标记物的质荷比。海洛因成瘾代谢组学研究具有一定的可行性,代谢组学在判定海洛因成瘾与否方面具有一定的潜在价值。
To establish an animal model of heroin addiction and study the changes of serum metabolites in heroin addicted rats and find out its potential markers. The experimental animals were divided into addicted group and blank control group. The effects of heroin addiction on the changes of serum metabolites in rats were studied by LC / MS-IT-TOF (liquid chromatography coupled with ion trap-time of flight mass spectrometry). Principal component analysis (PCA) and partial least-squares-discriminant analysis (PLS-DA) were used to analyze the metabolic differences between addicts and normal controls. The results showed that the addiction group and the control group had significant differences in serum metabolic level, and the mass-to-charge ratios of 26 potential biomarkers were initially screened out by statistical analysis. It is feasible to study the heroin addiction metabolomics. Metabolomics has some potential value in judging the addiction of heroin.
