生长激素释放抑制因子集中于肛门直肠及骨盆的神经中,似乎起着一种重要的神经递质作用。但是,既往分析奥曲肽(一种生长激素释放抑制因子的同类物)对肛门的作用则显示出相反的结果。通过采用在健康人群中建立的动力学模型,与高血糖素的肌原性作用作比较,其目的是为了进一步探索奥曲肽的药理学作用靶位。 方法:在一项安慰剂对照、随机、双盲交叉研究中,给12位健康志愿者分别在不同日期静脉滴注奥曲肽、高血糖素和安慰剂。在每一种序列中,以电子测压器用于测定直肠等压扩张的三种不同方案中的几种压力梯度,同时记录肛管的侧压反应、适合的容积及直肠感觉评分。
Ghrelin concentrations appear to concentrate on the anorectal and pelvic nerves and appear to play an important neurotransmitter role. However, previous analyzes of the effect of octreotide, a congener of the somatostatin, on the anus showed the opposite result. The aim of this study was to further explore the pharmacological target of octreotide by using the kinetic model established in healthy subjects in comparison with the myogenic nature of glucagon. Methods: In a placebo-controlled, randomized, double-blind crossover study, 12 healthy volunteers were dosed intravenously with octreotide, glucagon, and placebo on different days. In each sequence, electronic pressure-gauges were used to measure several pressure gradients in three different regimens of rectal isobaric dilation while recording the anal canal lateral pressure response, appropriate volume, and rectal sensation scores.
