为探讨尿激酶型纤溶酶原激活剂(uPA)及其抑制剂PAI-1mRNAs的表达变化与人肝细胞癌(HCC)浸润、转移间的关系,应用Northern印迹杂交技术对30例新鲜活检肝组织作uPA和PAI-1mRNAs表达的研究。结果显示:16例伴肝内浸润和转移癌组织中uPA和PAI-1mRNAs表达水平明显高于14例无肝内浸润和转移肝组织;18例高分化HCC癌组织与12例中、低分化HCC相比,uPA和PAI-1mRNAs含量差异有显著性意义。结果表明,癌组织内uPA和PAI-1基因转录水平的改变与HCC浸润、转移及细胞分化密切相关。
To investigate the relationship between the expression changes of urokinase-type plasminogen activator (uPA) and its inhibitor PAI-1 mRNAs and the invasion and metastasis of human hepatocellular carcinoma (HCC), Northern blot hybridization was used to detect 30 cases of fresh liver biopsy. Tissues were studied for the expression of uPA and PAI-1 mRNAs. The results showed that the expression levels of uPA and PAI-1 mRNAs in 16 cases with intrahepatic infiltration and metastatic carcinoma were significantly higher than those without intrahepatic infiltration and metastasis in 14 cases; 18 cases of well-differentiated HCC carcinoma and 12 cases of moderately-differentiated HCC In comparison, there were significant differences in the levels of uPA and PAI-1 mRNAs. The results showed that the changes of uPA and PAI-1 gene transcription levels in cancer tissues were closely related to infiltration, metastasis and cell differentiation of HCC.
