目的探讨慢性皮质酮(CORT)给药对CD-1小鼠行为学和海马CA1、CA3、齿状回(DG)亚区五羟色胺转载体(5-HTT)蛋白表达的影响。方法雄性CD-1小鼠皮下注射CORT(20 mg/kg,1次/d,连续3周)。每周检测皮毛状态、蔗糖偏好实验、强迫游泳实验(FST)和悬尾实验(TST),评估小鼠的行为学变化。3周后断头处死小鼠,进行海马分区,测定CA1、CA3、DG中5-HTT的蛋白表达。结果 CORT给药3周后,小鼠的皮毛状态退化,蔗糖偏好指数降低,FST和TST中小鼠的静止时间延长;海马3个亚区5-HTT的蛋白表达呈现亚区特异性下降,以DG亚区内的下调最为明显,其次为CA3,CA1内无明显变化。结论慢性CORT给药,诱导小鼠的抑郁样行为学表观,下调海马DG亚区5-HTT的蛋白表达。
Objective To investigate the effects of chronic corticosterone (CORT) administration on the behavior and the serotonin transporter (5-HTT) protein expression in the hippocampal CA1, CA3 and DG of CD-1 mice. Methods Male CD-1 mice were injected subcutaneously with CORT (20 mg / kg once daily for 3 weeks). Weekly measurements of fur status, sucrose preference test, forced swimming test (FST) and tail suspension test (TST) were used to assess behavioral changes in mice. Three weeks later, the mice were sacrificed and the hippocampus was divided into sections. The protein expression of 5-HTT in CA1, CA3 and DG was determined. Results After 3 weeks of administration of CORT, the fur status of mice was degraded, the index of sucrose preference was decreased and the resting time of mice in FST and TST was prolonged. The protein expression of 5-HTT in the three sub-regions of hippocampus showed a sub-region specific decrease. The most significant reduction in sub-region, followed by CA3, CA1 no significant change. Conclusions Chronic administration of CORT induces depression-like behavior in mice and down-regulates the protein expression of 5-HTT in DG subregion of hippocampus.
