目的探讨不同固定液和固定时间对大鼠睾丸组织病理学制片质量的影响。方法选用雄性SD大鼠12只,取两侧睾丸组织,共24例标本。每例标本分别用不同的固定液(10%中性福尔马林溶液、30%Davidson固定液及FAA固定液)固定不同的时间(24、48 h)后,进行脱水、包埋、切片、HE染色。用LEICA DM5000B普通光学显微镜观察。结果 3种固定液固定24 h后,可见30%Davidsons固定液组睾丸组织的异常变化程度低于10%中性福尔马林组和FAA组;固定时间对睾丸组织有一定程度的影响:固定24 h的异常变化程度低于固定48 h。结论 30%Davidson固定液组SD大鼠睾丸组织病理学变化程度最小;固定时间不宜超过24 h。
Objective To investigate the effects of different fixation solutions and fixed time on the histopathology of testis in rats. Methods Twelve male SD rats were selected, and the testes of both sides were taken for a total of 24 specimens. Each sample was fixed with different fixative solution (10% neutral formalin solution, 30% Davidson fixative solution and FAA fixative solution) for different time (24,48 h), dehydrated, embedded, HE staining. Using LEICA DM5000B ordinary optical microscope. Results After three kinds of fixatives were fixed for 24 h, the abnormal changes of testis in 30% Davidsons fixation group were lower than those in 10% neutral formalin group and FAA group. Fixation time had some effect on testicular tissue: 24 h abnormal changes less than the fixed 48 h. Conclusion The pathological changes of testis in 30% Davidson fixative group were the least. The fixation time should not exceed 24 hours.
