目的:探讨穿心莲内酯联合5-Fu对人低分化胃癌细胞株BGC-823的协同抗肿瘤作用。方法:以人胃癌BGC-823细胞株为靶细胞,通过体外细胞培养和裸鼠移植瘤试验,采用MTT法检测、绘制效应-合用指数曲线、抑瘤率计算等方法。结果:穿心莲内酯在7.5~120μg/mL、5-FU在3.75~60μg/mL范围内均能抑制BGC-823细胞的生长,具有剂量-效应关系和时间-效应关系。两药联合作用24h、48h、72h时的中效抑制浓度分别为17.32、9.54、4.11μg/mL,显著小于单药应用组。72h效应-合用指数曲线表明两药在抑制效应≤0.97范围内可产生协同作用。穿心莲内酯联合5-FU对人BGC-823细胞裸鼠移植瘤的抑瘤率为(75.51±12.57)%,与单药组相比差异均有显著性。结论:穿心莲内酯、5-Fu能在体内外抑制人胃癌BGC-823细胞的生长,两药联合产生协同或相加作用。
Objective: To investigate the synergistic antitumor effect of andrographolide combined with 5-Fu on human poorly differentiated gastric cancer cell line BGC-823. Methods: Human gastric cancer cell line BGC-823 was used as target cells. Cell culture and nude mouse xenograft in vitro were used to test the effect of MTT assay. The effect-combination index curve and tumor inhibition rate were calculated. RESULTS: Andrographolide inhibited the growth of BGC-823 cells in the range of 7.5 ~ 120μg / mL and 5-FU in the range of 3.75 ~ 60μg / mL with dose-effect relationship and time-effect relationship. The combined effect of two drugs 24h, 48h, 72h when the inhibitory concentrations were 17.32,9.54,4.11μg / mL, significantly less than the single drug application group. 72h effect - co-index curve shows that the two drugs in the inhibition effect ≤ 0.97 range can produce synergies. The inhibitory rate of andrographolide combined with 5-FU on human BGC-823 cell xenografts in nude mice was (75.51 ± 12.57)%, which was significantly different from that of monotherapy group. Conclusion: Andrographolide and 5-Fu can inhibit the growth of human gastric cancer cell line BGC-823 in vitro and in vivo, and the combination of the two drugs have a synergistic or additive effect.
