目的:优选聚酰胺树脂分离纯化绞股蓝总黄酮的工艺条件。方法:采用紫外分光光度法,以总黄酮含量为考察指标,研究在静态和动态条件下,聚酰胺树脂吸附和洗脱总黄酮的行为,筛选出聚酰胺树脂精制绞股蓝总黄酮的最优工艺参数。结果:最佳工艺参数为生药质量浓度0.35 g.mL-1,上样液pH 3,温度为室温,4 BV 60%乙醇洗脱。此工艺条件下,总黄酮纯度由粗品的23.91%提高到81.97%,纯化物中总黄酮保留率高达90.11%。结论:采用聚酰胺树脂纯化绞股蓝总黄酮效果良好,适用于工业化生产。
OBJECTIVE: To optimize the conditions for the separation and purification of total flavonoids from Gynostemma pentaphyllum with polyamide resin. Methods: Ultraviolet spectrophotometry was used to determine the total flavonoids content. Under static and dynamic conditions, the adsorption and elution of total flavonoids of polyamide resin were investigated. The optimum process parameters of total flavonoids of polyamide resin for gynostemma were screened out . Results: The optimum parameters were as follows: crude drug concentration 0.35 g.mL-1, sample solution pH 3, room temperature and 4 BV 60% ethanol. Under these conditions, the purity of total flavonoids increased from 23.91% of the crude product to 81.97%, and the retention of total flavonoids in the purified product reached 90.11%. Conclusion: Polygonatum resin is effective in purifying total flavonoids from Gynostemma pentaphyllum and is suitable for industrialized production.
