目的通过研究黄芪丹参1∶1配伍对缺氧缺血性脑损伤(HIBD)乳鼠脑组织Caspase-3、Bcl-2及Bax蛋白表达的影响,探讨其对脑组织的保护作用及机制。方法选择生后7 d SD乳鼠77只,随机分为假手术组(28只)、模型组(28只)及中药组(21只),应用免疫组化法观察不同时间点各组乳鼠脑组织Caspase-3、Bcl-2及Bax蛋白的表达情况。结果模型组在造模后各时间点Caspase-3及Bax的含量均显著高于假手术组,差异有统计学意义(P<0.05),模型组在造模2 h后Bcl-2含量显著高于假手术组,差异有统计学意义(P<0.05),在造模1 d、3 d后Bcl-2含量有升高趋势,但差异无统计学意义,造模7 d后,Bcl-2含量有下降趋势;中药组在造模后各时间点Caspase-3、Bax含量均较模型组显著降低,Bcl-2含量显著升高,差异有统计学意义(P<0.05)。结论黄芪丹参1∶1配伍能有效上调HIBD乳鼠脑组织Bcl-2蛋白表达,下调Caspase-3及Bax蛋白表达,从而抑制神经细胞凋亡,减轻缺氧缺血所致的脑组织损伤。
Objective To study the effects of Radix Salviae Miltiorrhizae 1: 1 mixture on the protein expression of Caspase-3, Bcl-2 and Bax in the brain of neonatal rats with hypoxic-ischemic brain damage (HIBD) and to explore its protective effect on brain. Methods Seventy-seven SD neonatal rats were selected and randomly divided into sham operation group (n = 28), model group (n = 28) and Chinese medicine group (n = 21). Immunohistochemical staining The expression of Caspase-3, Bcl-2 and Bax in brain tissue. Results The contents of Caspase-3 and Bax in the model group at each time point were significantly higher than those in the sham operation group (P <0.05). The content of Bcl-2 in the model group at 2 h after modeling was significantly higher In the sham group, the difference was statistically significant (P <0.05). After 1 and 3 days of modeling, the content of Bcl-2 increased, but the difference was not statistically significant. After 7 days, Bcl-2 (P <0.05). The contents of Caspase-3 and Bax in Chinese herbal medicine group were significantly lower than those in model group at each time point after modeling, and the content of Bcl-2 was significantly increased (P <0.05). Conclusion Radix Astragali and Radix Salviae miltiorrhizae 1: 1 can effectively up-regulate the expression of Bcl-2 protein and down-regulate the expression of Caspase-3 and Bax in brain tissue of neonatal HIBD neonatal rats, thus inhibiting neuronal apoptosis and decreasing the damage of brain tissue caused by hypoxia and ischemia.
