目的:研究人参皂苷Rh2(Ginsenoside Rh2)对人卵巢癌细胞的抑制作用,并初步探讨其抑制机制。方法:①人卵巢癌细胞SKOV3常规培养后,MTT法观察Rh2对SKOV3细胞增殖的抑制作用;②RT-PCR技术从分子水平检测SKOV3细胞Caspase3的mRNA表达,初步探讨其抑癌机制。结果:①MTT法检测结果显示:SKOV3细胞生长抑制率随Rh2浓度的增加而增大,与对照组比较,细胞增殖受到明显抑制(P<0.05)。②RT-PCR结果显示:实验组Caspase3的表达明显高于对照组。结论:①人参皂苷Rh2可抑制卵巢癌SKOV3细胞的增殖。②人参皂苷Rh2可能通上调caspase3的表达诱导卵巢细胞凋亡。
Objective: To study the inhibitory effect of ginsenoside Rh2 on human ovarian cancer cells and to investigate its inhibitory mechanism. Methods: (1) After SKOV3 human ovarian cancer cells were cultured routinely, MTT assay was used to observe the inhibitory effect of Rh2 on the proliferation of SKOV3 cells. ②RT-PCR was used to detect the mRNA expression of Caspase3 in SKOV3 cells at molecular level. Results: (1) The results of MTT assay showed that the growth inhibition rate of SKOV3 cells increased with the increase of Rh2 concentration, and the cell proliferation was significantly inhibited compared with the control group (P <0.05). ② RT-PCR results showed that: the experimental group Caspase3 expression was significantly higher than the control group. Conclusion: ① ginsenoside Rh2 can inhibit the proliferation of ovarian cancer SKOV3 cells. ② ginsenoside Rh2 may up-regulate the expression of caspase3 to induce ovarian cell apoptosis.
