目的:对艾烟中可吸入颗粒物冷凝液(MSC)进行鼠伤寒沙门氏菌回复突变试验,评价艾烟的潜在致突变性。方法:采用平板掺入法观察艾烟引发试验选用菌株TA97、TA98、TA100、TA102的回变菌落数。试验设0、62.5、125、250、500、1 000μg/皿,阳性对照组,分别做代谢系统活化及非活化的对照,每组设3个平行。在背景生长良好的条件下,计数受试物组的菌落回变数,回变菌落数等于或大于2乘以未处理对照数并有剂量反应关系或至少某一测试点有可重复的并有统计学意义的阳性反应,即可认为该受试物诱变试验阳性。结果:MSC在活化下TA98诱变阳性,而对TA97、TA100、TA102活化以及非活化、TA98非活化均为阴性,此结果可以重复并且差异有统计学意义。结论:MSC使细菌生长增多,并且有剂量反应关系,根据本实验结果MSC具有潜在的致基因突变性。
OBJECTIVE: To evaluate the mutagenicity of Aspergillus fumigatus by detecting the Salmonella typhimurium reversion of inhalable particulate condensate (MSC) in Aizhong tobacco. Methods: The mixed colonies of TA97, TA98, TA100, TA102 were used to observe the number of retrograde colonies. Experiment set 0,62.5,125,250,500,1 000μg / dish, the positive control group, respectively metabolic activation and non-activation of the control, each set 3 parallel. Under good background growth conditions, the number of colonies retriggered in the test group was counted, the number of colonies returned was equal to or greater than 2 times the number of untreated controls and there was a dose-response relationship or at least one test point was reproducible and statistically significant The significance of the positive reaction, that is, the test mutagenicity test positive. Results: MSCs were positive for TA98 mutagenesis but negative for TA97, TA100, TA102 activation and non-activation, TA98 non-activation. The results were repeatable and the difference was statistically significant. CONCLUSIONS: MSC increased bacterial growth and had a dose-response relationship. According to the results of this experiment, MSCs have potential mutagenicity.
