本实验旨在探讨家兔服利福平(RFP)对优降糖(Gli)药代动力学及降糖效应的影响。实验采用自身对照法。研究结果表明:Gli在家兔体内的药代动力学符合一室开放模型。RFP可减少Gli的吸收,加速Gli的代谢,显著改变Gli的药代动力学特征。文中列出实验所得Glita及RFP+Gli组Gli的主要药代参数。RFP对GIi的降糖作用在16h内产生增强效应,而在16h后消失。提示在该实验条件下,Gli可能有新的代谢产物生成,且有较强的降糖作用。
The purpose of this experiment was to investigate the effect of rabbit furosemide (RFP) on the pharmacokinetics and hypoglycemic effect of glyburide (Gli). Experiments using self-control method. The results show that: Gli in rabbits pharmacokinetics consistent with open-chamber model. RFP reduces the absorption of Gli, accelerates the metabolism of Gli and significantly changes the pharmacokinetic profile of Gli. The main pharmacokinetic parameters of Gli in experimental Glita and RFP + Gli groups are listed. The hypoglycemic effect of RFP on GIi produced an enhanced effect within 16 h, but disappeared after 16 h. It is suggested that under this experimental condition, Gli may produce new metabolites and has a strong hypoglycemic effect.
