目的:通过处方前研究,考察芍药苷的溶解特性,测定芍药苷在不同pH值缓冲液中的油水分配系数,为芍药苷脂质体的处方设计提供参考。方法:用振摇法测定芍药苷的溶解度及正辛醇-水分配系数,比较pH值对油水分配系数的影响。结果:芍药苷在水、氯仿中的溶解度分别为50.57mgmL-1、68.34mgmL-1;在水、pH值分别为2.0、2.5、4.0、5.0、5.8、6.8、7.8的磷酸盐缓冲液中的正辛醇-水分配系数分别为-0.47、-0.61、-0.76、-0.71、-0.50、-0.78、-0.98、-0.58。结论:芍药苷易溶于水和氯仿;pH对芍药苷的油水分配系数有一定的影响,在pH5.0缓冲盐溶液中的溶解度及油水分配系数最大。
OBJECTIVE: To study the dissolution characteristics of paeoniflorin by pre-prescription study and to determine the oil-water partition coefficient of paeoniflorin in different pH buffers to provide reference for the formulation design of paeoniflorin liposomes. Methods: The solubility of paeoniflorin and the partition coefficient of n-octanol-water were determined by shaking method. The effect of pH on the partition coefficients of oil and water was compared. Results: The solubility of paeoniflorin in water and chloroform was 50.57mgmL-1, 68.34mgmL-1, respectively. In water, the pH was 2.0,2.5,4.0,5.0,5.8,6.8,7.8 in phosphate buffer N-octanol - water partition coefficients were -0.47, -0.61, -0.76, -0.71, -0.50, -0.78, -0.98, -0.58. CONCLUSION: Paeoniflorin is soluble in water and chloroform. PH has some influence on the oil-water partition coefficient of paeoniflorin. The solubility and the partition coefficient of oil-water in pH 5.0 buffer solution are the highest.
