目的:建立适宜的2型糖尿病(T2DM)大鼠模型,在此基础上探讨T2DM并发高血压病的可能机制及中药复方的干预作用。方法:采用高脂高糖饲料喂养5周联合一次性腹腔注射30mg/kg链脲佐菌素方式造模,成模2周后大鼠分别给予盐酸吡格列酮(2.7mg/kg)和中药复方(3.8g/kg)灌胃,连续干预14d,测定相关实验室指标。结果:模型组大鼠空腹血糖、糖化血红蛋白、血清胰岛素、同型半胱氨酸、内皮素-1、收缩压水平、胰岛素抵抗指数、血栓素B2/6-酮前列腺素F1α比值均显著升高(P<0.01),胰岛素敏感指数、环磷酸腺苷/环磷酸鸟苷比值均显著下降(P<0.01);盐酸吡格列酮与中药复方两组大鼠的上述指标均显著改善(P<0.05,P<0.01)。结论:Hcy可能通过减弱内皮功能引发T2DM并发高血压病,中药复方对T2DM的血糖和血压具有调节作用。
OBJECTIVE: To establish a suitable rat model of type 2 diabetes mellitus (T2DM) and to explore the possible mechanism of T2DM complicated with hypertension and the intervention of traditional Chinese medicine. Methods: The rats were given pioglitazone hydrochloride (2.7mg / kg) and Chinese medicine compound (3.8mg / kg) for 2 weeks after being injected with 30mg / kg streptozotocin (STZ) g / kg) were given intragastrically for 14 days. The relevant laboratory parameters were measured. Results: The levels of fasting blood glucose, glycosylated hemoglobin, serum insulin, homocysteine, endothelin-1, systolic blood pressure, insulin resistance index and thromboxane B2 / 6-ketoprostaglandin F1α in model group were significantly increased P <0.01). The insulin sensitivity index and cyclic adenosine monophosphate / cyclic guanosine monophosphate ratio decreased significantly (P <0.01). The above indexes of pioglitazone hydrochloride group and traditional Chinese medicine compound group were significantly improved (P <0.05, P < 0.01). Conclusion: Hcy may induce T2DM complicated with hypertension by attenuating endothelial function. Chinese herbal compound has a regulatory effect on blood glucose and blood pressure of T2DM.
