目的对新疆阿勒泰牧区哈萨克传统发酵驼乳中分离出的一株乳酸菌进行分子生物学鉴定。方法采用DNA提取、PCR扩增、16SrDNA产物测序和数据分析对该菌株进行系统发育分析鉴定。结果 16SrDNA产物测序结果通过Eztaxon数据库比对,保守基因rpoA、pheS的扩增产物通过NCBI数据库比对,发酵驼乳中乳酸菌菌株的序列和已报道的乳酸菌菌株16SrDNA、rpoA、pheS的序列分别用MEGA 5.2软件进行系统发育分析表明:此菌株(10号)的16SrDNA序列与Lactobacillus pentosus(JCM 1558T)、Lactobacillus plantarumsubsp.Argentoratensis(DKO 22T)、Lactobacillus paraplantarum(DSM 10667T)和Lactobacillus plantarum subsp.Plantarum(ATCC 14917T)的相似率分别为100%、99.87%、99.8%和99.74%。结论 10号乳酸菌菌种经分子生物学鉴定为Lactobacillus pentosus(JCM 1558T)。
Objective To identify the molecular biology of a lactic acid bacterium isolated from the traditional Kazakh camel milk from the Altay pastoral area in Xinjiang. Methods The strain was identified by phylogenetic analysis using DNA extraction, PCR amplification, 16S rDNA sequencing and data analysis. Results The sequencing results of 16S rDNA were compared by Eztaxon database. The amplified products of conserved genes rpoA and pheS were compared by NCBI database. The sequences of lactobacillus in camel milk and 16S rDNA, rpoA and pheS of reported lactic acid bacteria strains were respectively sequenced by MEGA 5.2 Phylogenetic analysis of the software showed that the 16S rDNA sequence of this strain (No. 10) was identical to the Lactobacillus pentosus (JCM 1558T), Lactobacillus plantarum subsp. Argentoratensis (DKO 22T), Lactobacillus paraplantarum (DSM 10667T) and Lactobacillus plantarum subsp. Plantarum (ATCC 14917T) The similarities were 100%, 99.87%, 99.8% and 99.74% respectively. Conclusion Lactobacillus pentosus No. 10 (JCM 1558T) was identified by molecular biology.
