目的:建立辛夷药材(望春花)挥发油的含量测定及以桉油精为指标的气相色谱测定的质量分析方法。方法:应用《中国药典》的挥发油测定方法对望春花10个主流栽培品种的挥发油进行含量测定;应用气相色谱法对望春花10个主流栽培品种的桉油精进行含量测定。采用HP-5二苯基聚硅氧烷共聚物(0.25μm×0.32 mm×30 m)毛细管柱,FID检测器,载气为高纯度氮气,程序升温:初始温度80℃保留6 min,以20℃min-1的速率升温至160℃,保留2 min;再以60℃min-1的速率升温至220℃,保留5 min;进样口温度240℃,检测器温度240℃。结果:挥发油含量依次为3.24%,4.64%,6.12%,3.64%,5.48%,6.98%,3.64%,3.24%,7.68%,2.88%。气相色谱测定桉油精含量:桉油精的进样量在1.1~10.45μg(r=0.999 8)线性关系良好,平均回收率为100.02%,RSD 1.12%;含量依次为2.076%,4.212%,3.689%,0.104 1%,1.590%,3.523%,2.930%,1.940%,4.520%,1.494%。结论:根据望春花的挥发油及以气相色谱法桉油精的含量测定,显示不同栽培品种的望春花挥发油和桉油精的含量都有一定的差别,可用于望春花不同栽培品种质量评价和质量标准的制定。
OBJECTIVE: To establish a method for the determination of volatile oil in Radix et Rhizoma Rhei (var. Chinensis) and the mass spectrometric method for the determination of eucalyptol by gas chromatography. Methods: The volatile oil of “Chinese Pharmacopoeia” was used to determine the content of volatile oil in 10 mainstream cultivars of Viburnum chinense. The contents of eucalyptol in 10 mainstream varieties of Viburnum were determined by gas chromatography. HP-5 diphenyl polysiloxane copolymer (0.25μm × 0.32mm × 30m) capillary column, FID detector, the carrier gas is high purity nitrogen, the program temperature: the initial temperature of 80 ℃ for 6 min, with 20 The temperature was raised to 160 ℃ for 2 min. The temperature was raised to 220 ℃ at the rate of 60 ℃ min-1 for 5 min. The inlet temperature was 240 ℃ and the detector temperature was 240 ℃. Results: The volatile oil contents were 3.24%, 4.64%, 6.12%, 3.64%, 5.48%, 6.98%, 3.64%, 3.24%, 7.68%, 2.88%, respectively. Gas chromatographic determination of eucalyptol content: Eucalyptol injection volume in the 1.1 ~ 10.45μg (r = 0.999 8) good linear relationship, the average recovery was 100.02%, RSD 1.12%; content followed by 2.076%, 4.212% 3.689%, 0.104 1%, 1.590%, 3.523%, 2.930%, 1.940%, 4.520%, 1.494%. Conclusion: According to the determination of the essential oil of Viburnum and the content of eucalyptol by gas chromatography, the contents of Volatile Oil and Eucalyptol in different cultivars are different, which can be used to evaluate the quality and quality of different cultivars Standard formulation.
