背景:实验性脑缺血动物模型对研究脑卒中发病机制、治疗及康复评价具有重要意义。影响模型成功及稳定性因素为提高模型成功率提供实验依据。目的:探讨影响大鼠大脑中动脉局灶性脑缺血模型成功率的相关因素。设计:随机对照实验研究。地点和对象:实验地点:解放军济南军区总医院全军神经内科中心动物实验室。动物:雄性Wistar大鼠75只,体质量260~350g,共分5组(n=15):A组:颈外动脉结扎(externalcarotidarteryocclusion,ECAO)+翼腭动脉结扎(pterygopalarteryocclusion,PPAO)+聚乙烯醇栓线;B组:E-CAO+PPAO+硅胶栓线;C组:ECAO+聚乙烯醇栓线;D组:ECAO+硅胶栓线;E组:假手术组。干预:A和B组采用丝线结扎翼腭动脉,近心端结扎颈总动脉,血管夹夹闭颈总动脉远端,在颈总动脉上剪一小口,边松血管夹,边进线(A组用聚乙烯醇栓线,B组用硅胶栓线),至有阻力不能再进为止。C和D组不结扎翼腭动脉,其余步骤相同。E组手术和结扎动脉与缺血动物相同,但颈内动脉内不栓线。主要观察指标:动物模型出现率;梗死体积。结果:A,B组动物的模型出现率较C,D组高,模型成功动物栓线进入颅内长度多在7mm左右,而不成功动物则多在4mm以下。A,C组较B,D组神经缺损症状出现早,同一时间内的梗死体积显著增大。结论:翼腭动脉结扎与否是影响模型成功率高低
BACKGROUND: Experimental cerebral ischemia in animal models is of great importance for studying the pathogenesis, treatment and rehabilitation evaluation of stroke. Influencing model success and stability factors provide experimental basis for improving model success rate. Objective: To investigate the factors affecting the success rate of focal cerebral ischemia model in middle cerebral artery of rats. Design: Randomized controlled trial. Location and object: Experimental Location: PLA General Hospital of PLA Jinan Military Medical Center Animal Laboratory. Animals: 75 male Wistar rats weighing 260-350 g were divided into 5 groups (n = 15): Group A: external carotid artery occlusion (ECAO) + pterygopalarteryocclusion (PPAO) + polyethylene Group B: E-CAO + PPAO + silicone plug; Group C: ECAO + PVA plug; Group D: ECAO + silicone plug; Group E: sham-operated group. Intervention: Group A and group B were ligated with the pterygopalatine artery by silk thread, the common carotid artery was ligated proximally, the distal end of the common carotid artery was closed with a vascular clamp, and a small incision was made in the common carotid artery. Group with polyvinyl alcohol thread, B group with silicone plug), to the resistance can not further advance. Group C and D did not ligate the pterygopalatine artery, the rest of the procedure was the same. Group E surgery and ligation of arteries and ischemic animals the same, but no internal carotid artery embolization. MAIN OUTCOME MEASURES: Incidence of animal models; infarct volume. Results: The appearance rate of model animals in group A and group B was higher than that in group C and D, and the length of intracranial plug in animal model was more than 7mm, while that in unsuccessful animals was more than 4mm. A, C group than B, D group appeared early neurological deficit, infarct volume at the same time significantly increased.
